Selective oxidation of methylcytosine in single base pair mismatches by osmium complex

Abstract

We have reported a novel method for methylcytosine (M)-positive epigenotyping, in which an osmium ion site-specifically binds to M in a single strand or a bulge structure of DNA on oxidation of an M C5-C6 double bond. Herein, we have examined the osmium oxidation reactivity of M that formed a mismatched base pair with various bases, and observed its unique reactivity.
